Phosphatidylinositol and Related Lipids

چکیده

Phosphatidylinositol is an important lipid, both as a key membrane constituent and as a participant in essential metabolic processes in all plants and animals, both directly and via a number of metabolites. It is an acidic (anionic) phospholipid that in essence consists of a phosphatidic acid backbone, linked via the phosphate group to inositol (hexahydroxycyclohexane). In most organisms, the stereochemical form of the latter is myo-D-inositol (with one axial hydroxyl in position 2 with the remainder equatorial), although other forms (scylloand chiro) have been found on occasion in plants. The 1-stearoyl,2-arachidonoyl molecular species, which is of considerable biological importance in animals, is illustrated.

GhLTPG1, a cotton GPI-anchored lipid transfer protein, regulates the transport of phosphatidylinositol monophosphates and cotton fiber elongation

The cotton fibers are seed trichomes that elongate from the ovule epidermis. Polar lipids are required for the quick enlargement of cell membrane and fiber cell growth, however, how lipids are transported from the ovules into the developing fibers remains less known. Here, we reported the functional characterization of GhLTPG1, a GPI-anchored lipid transport protein, during cotton fiber elongat...

Modulation of the in vitro activity of lysosomal phospholipase A1 by membrane lipids.

Lysosomal phospholipases play a critical role for degradation of cellular membranes after their lysosomal segregation. We investigated the regulation of lysosomal phospholipase A1 by cholesterol, phosphatidylethanolamine, and negatively-charged lipids in correlation with changes of biophysical properties of the membranes induced by these lipids. Lysosomal phospholipase A1 activity was determine...

Corticotropin-(1--24)-tetracosapeptide affects protein phosphorylation and polyphosphoinositide metabolism in rat brain.

1. Effects of corticotropin-(1--24)-tetracosapeptide on the endogenous phosphorylation of proteins and lipids were studied in a membrane/cytosol fraction prepared from a lysed crude mitochondrial/synaptosomal fraction. 2. The labelling of proteins and lipids was monitored by incubation of the subcellular fraction for 10s with [gamma-32P]ATP. 3. The phosphorylation of proteins was dose-dependent...

Analysis of the metabolic turnover of the individual phosphate groups of phosphatidylinositol 4 - phosphate and phosphatidylinositol 4 , 5 - bisphosphate

1. We have developed methods that yield estimates of the 32p content of each of the individual phosphate groups of phosphatidylinositol 4-phosphate and phosphatidylinositol 4,5-bisphosphate, thus extending the information available from studies of the labelling of these lipids in intact cells or membrane preparations. The analyses are undertaken with the deacylated lipids. 2. Assay of the 5-pho...

Intracellular Signalling: New directions for phosphatidylinositol transfer

Newly revealed properties of phosphatidylinositol transfer protein help to explain the cellular targeting of lipids involved in signal transduction, and indicate that inositol lipids play a part in directing membrane traffic.

Metabolism of fatty acids by ovine spermatozoa.

The incorporation of (14)C-labelled myristic, palmitic, stearic, oleic and linoleic acids in vitro into the lipids of bovine spermatozoa was measured at intervals from 2min to 2h. All acids were rapidly incorporated into diglycerides, myristic acid being metabolized to the greatest extent. Whereas the low incorporation of acids into total phospholipids reflected the relative stability of the ma...
